Triple negative breast cancer (TNBC) is a form of BC characterized by high aggressiveness, therapy resistance, short time to relapse, poor prognosis. The presence of Cancer Stem Cells (CSCs) could be responsible for TNBC resistance to therapy, recurrence and metastasis, and might explain the difficult of its eradication. Mcl-1 is one of the key regulators of CSCs self-renewal and its expression can limit the efficacy of antitumorigenic agents as TRAIL, a selective anticancer agent but with limited effects against some cancer cell lines. Triple negative breast cancers (TNBCs), characterized by lack of estrogen, progesterone and HER2 receptors, are a highly heterogenous group of tumors which account for about 20% to 25% of all breast cancers. TNBCs are often associated with epithelial-mesenchymal transition and a high propensity for early metastasis. Since no molecularly-targeted therapeutic agents are clinically available for TNBCs, these tumors, which are frequently resistant to cytotoxic chemotherapy, remain difficult to treat. Nevertheless, progress is being made in the finding of molecular alterations typical of TNBCs toward which to focus therapeutic efforts.

The HDAC inhibitor SAHA synergistically stimulates the cytotoxic effect induced by Parthenolide in MDA-MB231 cells

We showed that the sesquiterpene lactone Parthenolide (PN) exerts strong cytotoxic effects on triple negative breast cancer MDA-MB231 cells. Our recent results suggest that PN exerts in these cells a cytoprotective effect, which is due to the activation of mTOR pathway. To inhibit this protective response we employ the HDAC inhibitor SAHA, which is known to prevent AKT/mTOR pathway. We show that PN activates Akt, mTOR, p70S6kinase and NRF2 while SAHA abolishes these effects. Further cell pretreatment with SAHA synergistically sensitizes the cells to the cytotoxic effect of PN. An amphiphilic inulin-thiocholesterol conjugate (INU-Cys-TC) was strategically designed as a biodegradable core-shell nanocarrier of 7-ethyl-10-hydroxy-camptothecin (SN38) to enhance its solubility and stability in aqueous media, thus exploiting its brilliant anticancer effect. INU-Cys-TC was designed to have the hydrophilic inulin backbone (external shell) partially functionalized with hydrophobic thiocholesterol moieties (internal core) through a biodegradable disulfide bond due to cysteamine bridges.
